•  bace1（β-site app cleaving enzyme），即β-分泌酶，是淀粉樣前體蛋白（amyloid precursor protein，app）產(chǎn)生aβ的關鍵酶。bace1屬于ⅰ型跨膜蛋白，由501個氨基酸組成，其中46～460位氨基酸為功能活性區(qū)域。bace1通過水解app670met-671asp之間的肽鍵，生成670個氨基酸的可溶性n末端和包含完整aβ區(qū)的app跨膜c端。后者經(jīng)γ分泌酶進一步酶切形成aβ。zui近的研究表明，bace1基因敲除小鼠腦內的aβ水平可降低90%，且其生理表型基本正常，顯示了bace1抑制劑在ad治療中的廣闊前景［1］。通過酵母或昆蟲表達系統(tǒng)表達活性bace1在zui近已見報道［2，3］。然而，由于上述系統(tǒng)表達的蛋白量低，且純化流程復雜，不適合bace1抑制劑的大規(guī)模篩選。而bace1的原核表達可克服以上缺陷，為篩選特異性抑制劑及研究bace1結構與功能關系提供大量材料，為zui終從根本上治療ad帶來希望。
